One great program that is commonly overlooked is Hijackthis, it is extremely beneficial. But someone who doesn't know what they are doing can mess up their computer.

Spybot S&D is a good program but i don't know why but i don't choose to have it as one of my spyware/adware remover
I myself only have ad-aware and although i recommend microsoft anti-spyware, i don't use it
The sp2 firewall takes care of most hijackers, viruses, etc
